应用逆转录聚合酶链反应(RT-PCR)技术,对C.jacchus猴实验性过敏性脑脊髓炎(EAE)脑组织中肿瘤坏死因子α(TNFα)mRNA的水平进行了研究。结果表明:EAE动物脑组织TNFαmRNA明显高于正常动物;用一种TNFα释放抑制剂环戊苯吡酮(rolipram)可阻断免疫动物EAE的发生和脑组织中TNFαmRNA水平降低。说明TNFα在EAE发生过程中具重要作用。采用抑制TNFα的手段,为多发性硬化症及相关疾病的治疗提供了新线索。
The level of tumor necrosis factor α (TNFα) mRNA in brain tissue of C.jacchus monkey experimental allergic encephalomyelitis (EAE) was studied by reverse transcription-polymerase chain reaction (RT-PCR). The results showed that TNFαmRNA in brain tissue of EAE animals was significantly higher than that of normal animals. The rolipram, a TNFα inhibitor, could block the EAE and decrease the level of TNFαmRNA in brain tissues. TNFα in the process of EAE play an important role. Suppression of TNFα means for the treatment of multiple sclerosis and related diseases provide new clues.
