目的:检测Jagged-1 mRNA和蛋白在乳腺癌及对照正常乳腺组织中的表达状况,分析Jagged-1的表达与乳腺癌临床病理特征的关系。方法:应用逆转录聚合酶链反应(RT-PCR)和免疫组化法检测Jagged-1 mRNA和蛋白质在60例乳腺癌和25例癌旁正常乳腺组织中的表达,统计分析其表达水平与乳腺癌临床病理参数间的联系。结果:Jagged-1蛋白阳性表达在乳腺癌中显著高于癌旁正常乳腺组织(P<0.05)。Jagged-1 mRNA阳性表达在乳腺癌组织TNM不同分期中有差异(P<0.05)。Jagged-1蛋白阳性表达在乳腺癌淋巴结转移组、Ⅱ和Ⅲ期临床分组、Ⅱ和Ⅲ级组织学分组分别显著高于无淋巴结转移组、0~Ⅰ期组和Ⅰ级组(P<0.05)。结论:Jagged-1 mRNA和蛋白表达在乳腺癌的发生和进展过程中起重要作用。
OBJECTIVE: To detect the expression of Jagged-1 mRNA and protein in breast cancer and normal breast tissues, and to analyze the relationship between Jagged-1 expression and the clinicopathological features of breast cancer. Methods: The expression of Jagged-1 mRNA and protein in 60 cases of breast cancer and 25 cases of adjacent normal breast tissue was detected by RT-PCR and immunohistochemistry. Results: The positive expression of Jagged-1 protein in breast cancer was significantly higher than that in normal breast tissues (P <0.05). The positive expression of Jagged-1 mRNA was different in different stages of TNM (P <0.05). The positive expression of Jagged-1 protein in breast cancer lymph node metastasis group Ⅱ and Ⅲ clinical group, Ⅱ and Ⅲ histological group were significantly higher than those without lymph node metastasis, 0 ~ Ⅰ group and Ⅰ group (P <0.05) . Conclusion: The expression of Jagged-1 mRNA and protein plays an important role in the carcinogenesis and progression of breast cancer.
