目的:考察动物种属差异及黏膜部位对药物经鼻黏膜渗透性的影响。方法:灯盏乙素为模型药物,采用立式扩散池,分别以新鲜的离体犬、小型猪、家猪离体鼻黏膜为渗透屏障,药物置于鼻黏膜上方,以pH6.8 PBS为接收液,进行渗透试验(n=4),HPLC法测定灯盏乙素。结果:灯盏乙素经犬、小型猪、家猪鼻黏膜渗透系数分别为(16.162±1.957)×10-3cm/min,(13.697±0.605)×10-3cm/min,(4.740±0.211)×10-3cm/min;灯盏乙素经上述动物离体鼻黏膜的渗透系数之间均存在显著性差异。灯盏乙素经离体家猪鼻中膈前部与鼻中膈黏膜的渗透系数分别为(0.194±0.004)×10-3cm/min、(4.740±0.211)×10-3cm/min,两者比较,具有显著性差异。结论:灯盏乙素可经上述离体鼻黏膜渗透,但渗透系数存在种属差异;灯盏乙素经同一动物鼻黏膜不同部位的渗透性亦存在差异。
Objective: To investigate the differences of animal species and mucosal parts of the nasal mucosa permeability of drugs. Methods: Scutellarin was used as a model drug. Vertical diffusion cells were used. Fresh nasal mucosa of isolated canine, miniature pig and domestic pig were used as osmotic barrier. The drug was placed on the nasal mucosa and received at pH 6.8 PBS Liquid, the penetration test (n = 4), determination of Scutellarin HPLC. Results: The osmotic coefficients of scutellarin in dogs, piglets and domestic pigs were (16.162 ± 1.957) × 10-3cm / min, (13.697 ± 0.605) × 10-3cm / min, (4.740 ± 0.211) × 10- 3cm / min; scutellarin nasal mucosa osmotic permeability coefficient between the animals there are significant differences. The osmotic coefficients of the scutellarin in the frontal part of nasal septum and nasal septum were (0.194 ± 0.004) × 10-3cm / min and (4.740 ± 0.211) × 10-3cm / min, respectively There are significant differences. CONCLUSIONS: Scutellarin is permeable to the nasal mucosa in vitro, but the osmotic coefficient is different in species. Scumbellin also has different permeability in different parts of the same animal’s nasal mucosa.
