目的研究佐剂关节炎(AA)大鼠心肺功能变化与NF-κBp65/TNF-α、TGF-β1/Smad通路的关系。方法将30只大鼠随机均分为正常对照(NC)组和模型对照(MC)组,向MC组大鼠右后足跖皮内注射弗氏完全佐剂0.1 ml致炎,复制成AA模型。致炎19 d后,观察大鼠足跖肿胀度、关节炎指数(AI),采用超声诊断仪检测大鼠心功能、小动物肺功能仪检测肺功能变化,免疫印迹法检测大鼠心肺NF-κBp65、TNF-α、TGF-β1、Smad2、Smad4、Smad7蛋白表达。结果与正常组比较,AA大鼠足趾肿胀,AI升高,心肺功能参数降低,心肺组织NF-κBp65、TNF-α、TGF-β1、Smad2、Smad4蛋白表达升高,Smad7降低(P<0.01或P<0.05);相关性分析显示,AA大鼠心肺功能参数与NF-κBp65/TNF-α、TGF-β1/Smad通路存在关联。结论 AA大鼠心肺功能损伤可能与NF-κBp65/TNF-α、TGF-β1/Smad通路过度激活有关。
Objective To investigate the relationship between changes of cardiopulmonary function and the pathways of NF-κBp65 / TNF-α and TGF-β1 / Smad in adjuvant arthritis (AA) rats. Methods Thirty rats were randomly divided into normal control group (NC) and model control group (MC). MCF rats were injected intraperitoneally with 0.1 ml Freund’s complete adjuvant intradermally to make inflammation in AA rats . After 19 days of inflammation, the degree of paw edema, the arthritis index (AI), the cardiac function of rats were detected by using the ultrasonic diagnostic apparatus, the changes of lung function were detected by the pulmonary function test of small animals, the expressions of NF- κBp65, TNF-α, TGF-β1, Smad2, Smad4, Smad7 protein expression. Results Compared with the normal control group, the toe swelling and AI in the AA rats were decreased, the cardiopulmonary function parameters were decreased, the expression of NF-κBp65, TNF-α, TGF-β1, Smad2 and Smad4 in cardiac and pulmonary tissues were increased and Smad7 was decreased Or P <0.05). Correlation analysis showed that the cardiopulmonary parameters of AA rats correlated with the NF-κBp65 / TNF-α and TGF-β1 / Smad pathways. Conclusion The cardiopulmonary dysfunction in AA rats may be related to the over-activation of NF-κBp65 / TNF-α and TGF-β1 / Smad pathways.
