目的分析NOTCH-1信号通路对人脑胶质瘤干细胞增殖和分化作用。方法选取医院2015年1月-2017年1月收治的人脑胶质瘤患者50例,从人脑胶质瘤组织标本和细胞株中提出胶质瘤干细胞体外培养,对胶质瘤干细胞增殖和分化作用中NOTCH-1信号通路基因和蛋白表达情况进行分析。结果高度恶性胶质瘤光密度值(IOD)高于低度恶性胶质瘤(P<0.05);在胶质瘤干细胞增殖过程中,NOTCH-1、CBF-1、HES-1在CD133+中表达较强,干细胞分化时,表达减弱。结论 NOTCH-1信号通路参与了人脑胶质瘤干细胞增殖和分化过程,在人脑胶质瘤中高表达,为疾病治疗提供了重要依据。
Objective To analyze the effect of NOTCH-1 signaling pathway on the proliferation and differentiation of human glioma stem cells. Methods Fifty patients with glioma admitted to our hospital from January 2015 to January 2017 were enrolled in this study. Glioma stem cells were cultured in vitro from human glioma tissue samples and cell lines. Differentiation in the NOTCH-1 signaling pathway gene and protein expression analysis. Results The IOD of high grade glioma was higher than that of low grade glioma (P <0.05). In the process of glioma stem cell proliferation, NOTCH-1, CBF-1 and HES-1 were expressed in CD133 + Strong, stem cell differentiation, the expression weakened. Conclusion The NOTCH-1 signaling pathway is involved in the proliferation and differentiation of human glioma stem cells and is highly expressed in human gliomas, providing an important basis for disease treatment.
