为了研究用温阳及补气两种温热药治疗虚寒证大鼠后,其脑及肾上腺内5—羟色胺(5—HT)及去甲肾上腺素(NE)神经元以及垂体内神经纤维的递质含量变化,采用免疫组织化学的方法进行神经元及纤维形态学观察及定量分析,用荧光测定法测量脑内两种递质的含量。结果温阳与补气药均使全脑的5—HT细胞及纤维着色变浅,NE细胞及纤维的染色加深。垂体内NE纤维递质含量增多,5—HT递质含量减少。肾上腺髓质内变化不明显。全脑的NE含量增加而5—HT的含量减少。说明温阳、补气药均能提高脑各区NE神经元递质的合成释放,并抑制5—HT的合成释放,两类药物作用效果无明显差别,上述的形态学改变与机能变化一致。
In order to study the effects of Wenyang and qi supplements on the treatment of Deficiency-Induced Cold Syndrome Rats, their brain and adrenal 5-hydroxytryptamine (5-HT) and norepinephrine (NE) neurons and pituitary nerve fibers Changes in neurotransmitter content were measured by immunohistochemical methods for neuronal and fiber morphology and quantitative analysis. The content of both neurotransmitters in the brain was measured by fluorescence measurement. RESULTS: Both Wenyang and qi-invigorating drugs decreased the staining of 5-HT cells and fibers in the whole brain, and the staining of NE cells and fibers increased. Pituitary NE content of neurotransmitters increased, 5-HT transmitter content decreased. Adrenal medulla changes are not obvious. Whole brain NE content increases and 5-HT content decreases. The results indicated that both Wenyang and qi can increase the synthesis and release of neurotransmitters in brain neurons and inhibit the synthesis and release of 5-HT. The effects of the two drugs have no significant difference. The above morphological changes are consistent with the changes of function.
