目的:通过测定大鼠心肌细胞ATP酶活性和Ca2+浓度来探讨补脾方药对脾虚证的作用机制。方法:SPF大鼠采用复合方法制成脾虚模型后分别以3种不同配伍补脾方药灌胃3周。采用比色分析法测定心肌细胞ATP酶活性和Ca2+浓度。结果:与正常对照组、模型组比较,中药用药组大鼠心肌细胞ATP酶活性显著升高(P<0.05),中药用药组大鼠心肌细胞Ca2+浓度比模型组有所降低(P<0.05),有统计学意义。结论:补脾方药治疗脾虚证的作用机制可能与提高细胞内ATP酶活性和降低Ca2+浓度有关。
Objective: To investigate the mechanism of invigorating the spleen recipe on spleen deficiency syndrome by measuring ATPase activity and Ca2 + concentration in rat cardiomyocytes. Methods: SPF rats were made into spleen deficiency model by the compound method and were respectively fed with 3 kinds of prescriptions of invigorating spleen and invigorating the spleen for 3 weeks. Cardiomyocyte ATPase activity and Ca2 + concentration were determined by colorimetric assay. Results: Compared with the normal control group and the model group, the ATPase activity of cardiomyocytes in the TCM-treated group was significantly increased (P <0.05), while the Ca2 + concentration in the cardiomyocytes of the TCM-treated group was lower than that in the model group (P <0.05) , With statistical significance. Conclusion: The mechanism of action of invigorating the spleen recipe on spleen deficiency syndrome may be related to the increase of intracellular ATPase activity and the decrease of Ca2 + concentration.
