目的:探讨肿瘤坏死因子-α(TNF-α)与葡萄膜炎发病的相关性。方法:采用多聚酶链反应技术检测60例葡萄膜炎及60例正常人外周血TNF-α水平及其基因多态性。结果:观察组外周血中TNF-α水平明显高于对照组(P<0.05);观察组经PCR扩增后可见特异性TNF-α扩增产物,TNF-α基因目的片段大小为117 b;对照组未见特异性TNF-α扩增产物;经PCR/RFLP分析发现两组TNF-α基因有3种酶切格局,即TNFα-308A/A纯合子的标本不发生酶切、TNFct-308G/G纯合子发生完全酶切;TNFα-308G/A杂合子产生部分酶切;两组基因型频率从高到低依次为G/G型、G/A型、A/A型,但两组比较具有明显差异(P<0.05)。结论:肿瘤坏死因子TNF-α与葡萄膜炎的发病具有显著的相关性。
Objective: To investigate the relationship between tumor necrosis factor-α (TNF-α) and the pathogenesis of uveitis. Methods: Polymerase chain reaction (PCR) was used to detect the levels of TNF-α in 60 patients with uveitis and 60 healthy controls and their genetic polymorphisms. Results: The level of TNF-α in the peripheral blood of the observation group was significantly higher than that of the control group (P <0.05). The specific amplification product of TNF-α was observed in the observation group after PCR amplification. The size of the target fragment of TNF- No specific TNF-α amplification product was found in the control group. Three restriction enzyme digestion patterns of TNF-α gene in the two groups were found by PCR / RFLP analysis. The TNFα-308A / / G homozygotes completely digested; TNFα-308G / A heterozygote partial digestion; two genotypes from high to low in order of G / G type, G / A type, A / A type, but the two groups Compared with the significant difference (P <0.05). Conclusion: There is a significant correlation between tumor necrosis factor-α and uveitis.
