目的 :观察α_半乳糖苷酶对猕猴类人B抗原的酶解效果及α_半乳糖苷酶酶解后的红细胞进行异种血型动物输血的安全性。方法 :用 10 0U/mlα_半乳糖苷酶酶解猕猴红细胞表面的类人B血型抗原 ,吸收放散法检测猕猴血型及α_半乳糖苷酶酶解后猕猴红细胞类人B血型抗原清除情况 ;同时检测酶解对红细胞结构与功能的影响 ;回输实验检测红细胞存活率以及观察酶解后红细胞的安全性。结果 :α_半乳糖苷酶可以清除猕猴红细胞表面类人B抗原 ,而不影响红细胞结构、功能及其在体内存活 ;α_半乳糖苷酶酶解的类人B型红细胞输给类人A型血的猕猴 ,未发生输血反应 ;受血猴的血液生化及尿液各项指标与输血前相比 ,无明显变化。结论 :α_半乳糖苷酶可以清除猕猴红细胞表面类人B抗原 ,酶解后的红细胞输给类人A型猕猴是安全的。
OBJECTIVE: To observe the effect of α-galactosidase on the enzymatic hydrolysis of cynomolgus human B antigen and the safety of blood transfusion of heterologous blood of red blood cells after α-galactosidase enzymolysis. Methods: Human erythrocyte human B blood group antigens were digested with 10 0 U / ml α-galactosidase, and the clearance of macaque erythrocyte human B blood group antigens by macaque blood group and α-galactosidase digestion was detected by absorption and emission assay. At the same time, the effect of enzymolysis on the structure and function of erythrocytes was also tested. The survival rate of erythrocytes and the safety of erythrocytes after enzymolysis were tested by back infusion experiment. Results: α-galactosidase could clear human erythrocyte surface human B antigen without affecting the structure and function of erythrocyte and its survival in vivo. Human α-galactosidase-like human B-type erythrocytes Blood-type rhesus monkeys, no transfusion reaction; blood monkey blood biochemical and urine indicators compared with before transfusion, no significant change. CONCLUSION: Alpha-galactosidase can eliminate rhesus erythrocyte surface humanoid B antigen, and the red blood cells after enzyme digestion are safe to human type A rhesus monkey.
