为探讨诱生型一氧化氮合酶 (iNOS)和内皮素 - 1(ET - 1)在糖尿病 (DM )大鼠早期肾组织中的动态表达及意义。利用链脲佐菌素 (STZ)复制糖尿病动物模型 ,免疫组化及计算机图像分析定位、半定量检测iNOS和ET - 1在肾组织中的表达。结果显示 :在DM大鼠肾组织中 ,iNOS以第 2周表达最多而第 4周表达下调 ,对照组中未见表达 ;ET - 1在正常大鼠及DM大鼠肾组织中均有表达 ,髓质多于皮质 ,DM大鼠肾小球中第1,2周较对照组明显降低 ,第 4周却升高 ,在肾小管 -间质区随病程延长而逐渐增加。由此提示NO、ET参与了DM早期肾小球高灌注和肾小管 -间质的损伤。
To investigate the dynamic expression of inducible nitric oxide synthase (iNOS) and endothelin - 1 (ET - 1) in the early stage of diabetic rat diabetic nephropathy. Animal models of diabetes mellitus were established by streptozotocin (STZ), immunohistochemistry and computer image analysis were performed to detect the expression of iNOS and ET - 1 in renal tissues. The results showed that the expression of iNOS was the highest in the second week and the fourth week in the DM rats, but not in the control group. The expression of ET - 1 was detected in the kidney of normal rats and DM rats, Medulla more than cortex, glomerular glomeruli in DM rats in the first and second weeks than in the control group was significantly lower, the fourth week but increased in the tubulointerstitial area with the course of the disease gradually increased. This suggests that NO, ET involved in early DM glomerular perfusion and tubulointerstitial injury.
