目的:探讨何首乌饮对衰老大鼠卵巢组织凋亡相关蛋白Bax/Bcl-2及Caspase-3表达的影响。方法:D-半乳糖连续腹腔注射致亚急性衰老动物模型,造模后灌胃给予何首乌饮,连续60 d,大鼠断头取卵巢。采用免疫组织化学分析方法检测各组大鼠凋亡相关基因Bcl-2和Bax的表达;Western blotting法检测各组大鼠卵巢Caspase-3蛋白表达的变化。结果:模型组较正常组卵巢细胞Bax和Caspase-3表达量增加,Bcl-2表达减弱;何首乌饮可使卵巢Bax和Caspase-3表达量降低,Bcl-2表达增加,并呈现一定的剂量依赖性,差异有统计学意义(P<0.05)。结论:何首乌饮方剂能延缓衰老大鼠卵巢细胞凋亡的发生,其机制可能是通过干预凋亡相关蛋白Bax/Bcl-2及Caspase-3的表达抑制卵巢细胞的凋亡。
Objective: To investigate the effect of Heshouwuyin on the expression of Bax / Bcl-2 and Caspase-3 in ovarian tissue of aging rats. Methods: The animal model of subacute aging was induced by continuous intraperitoneal injection of D-galactose. After the model was given, the Polygonum multiflorum Thunb drink was administered intragastrically for 60 days. The rats were decapitated and ovariectomized. Immunohistochemistry was used to detect the expression of Bcl-2 and Bax in each group, and Western blotting was used to detect the expression of Caspase-3 in ovaries. Results: Compared with the normal group, the expression of Bax and Caspase-3 in the model group increased, while the expression of Bcl-2 decreased in the model group. Polygonum multiflorum Decoction could reduce the expression of Bax and Caspase-3 in ovary, and increase the expression of Bcl-2 in a dose-dependent manner Sex, the difference was statistically significant (P <0.05). Conclusion: The recipe of Polygonum multiflorum Decoction can delay the apoptosis of ovarian cells in aging rats. The mechanism may be that the apoptosis of ovarian cells may be inhibited by the intervention of Bax / Bcl-2 and Caspase-3.
