目的 观察丹参酸乙对大鼠肝星状细胞α-平滑肌肌动蛋白的影响。方法 原位灌注、消化法分离正常大鼠肝星状细胞,异硫氰酸胍一步法提取细胞总RNA,RT-PCR法检测α-平滑肌肌动蛋白基因表达,免疫印迹法分析α-平滑肌肌动蛋白量。结果 10μmol/L SAB可抑制肝星状细胞α-平滑肌肌动蛋白的基因表达,但不能抑制其蛋白表达。细胞经TGFβ1刺激后,不仅1μmol/L SAB甚至10μmol/L SAB均不能抑制a-平滑肌肌动蛋白的基因表达。结论 丹参酸乙在肝星状细胞活化的启动阶段可延缓其活化,对于已活化的细胞丹参酸乙则无逆转作用。
Objective To observe the effects of salvianic acid B on α-smooth muscle actin in rat hepatic stellate cells. METHODS: Normal rat hepatic stellate cells were isolated by in situ perfusion and digestion. Total cellular RNA was extracted by guanidinium thiocyanate method. α-smooth muscle actin gene expression was detected by RT-PCR. α-smooth muscle was analyzed by Western blotting. The amount of protein. Results 10μmol/L SAB could inhibit the gene expression of α-smooth muscle actin in hepatic stellate cells, but it could not inhibit the protein expression. After stimulated by TGFβ1, not only 1μmol/L SAB but also 10μmol/L SAB could not inhibit a-smooth muscle actin gene expression. Conclusion Salvianic acid B can delay the activation of hepatic stellate cells in the activation phase, but it has no reversal effect on the activated cells.
