对数期生长的细菌代谢较旺盛,其DNA在复制过程中易受致突变物的影响。据此,本文比较了旋转培养(60转/分)5小时和16小时Ames检试菌TA1535、TA100、TA98、TA37,分别对叠氮钠、苦酮酸、硝基腐植酸的致突变效应。经多次实验结果表明:旋转培养5小时的细菌受相应的致突变物诱发的回变菌落数/平板,与旋转培养16小时者相比,均有显著增加,其回变菌落数的差异经统计学处理,大都具有显著或高度显著意义。即旋转培养5小时的细菌对致突变物的敏感性有所增强。从检试菌的活菌数和自发回变菌落数来看,培养时间也不宜过长。
Bacterial growth during logarithmic phase is exuberant and its DNA is susceptible to mutagenic effects during replication. Therefore, we compared the mutagenic effects of Ames test strains TA1535, TA100, TA98, TA37 at 5h and 16h with sodium azide, picoric acid and nitro humic acid respectively. The results of multiple experiments showed that the number of colonies returned by the corresponding mutagen-induced bacteria was significantly increased after cultured for 5 hours. The number of colonies recovered after 16 hours of rotation was significantly increased Statistical treatment, mostly with significant or highly significant. That is, the bacteria that are cultured for 5 hours by rotation are more susceptible to mutagens. From the test of viable bacteria and spontaneous change back to the number of colonies, culture time should not be too long.
