在培养的肝星状细胞系中加入不同浓度的重组人肝再生增强因子 (hALR) ,于不同的时间点收集细胞 ,提取总RNA ,用逆转录定量PCR方法测定Ⅰ、Ⅲ型胶原的基因表达水平。结果表明 ,高 ( 0 2ng/L)、中( 0 0 2ng/L)、低 ( 0 0 0 2ng/L)浓度的hALR 3组肝星状细胞Ⅰ型胶原基因表达水平在 8、2 4、48、72h 4个时间点均明显低于对照组 ;大剂量组较中、低剂量组Ⅰ型胶原基因表达水平亦明显为低。中、低剂量hALR组肝星状细胞Ⅲ型胶原基因表达水平在 2 4、48、72h 3个时间点明显低于对照组 ;大剂量hALR组肝星状细胞Ⅲ型胶原基因表达水平在 8、2 4、48、72h 4个时间点均明显低于对照组 ,较中、小剂量组亦显著降低。提示重组人肝再生增强因子对肝星状细胞Ⅰ、Ⅲ型胶原基因表达有明显的抑制作用。
Different concentrations of recombinant human augmenter of liver regeneration (hALR) were added into the cultured hepatic stellate cell line, and the cells were collected at different time points to extract the total RNA. The mRNA expression of type Ⅰ and type Ⅲ collagen was determined by reverse transcription quantitative PCR Level. The results showed that the gene expression of type Ⅰ collagen in hepatic stellate cells of high (0 2ng / L), medium (0 2ng / L) and low (0 0 0 2ng / L) 48, 72h four time points were significantly lower than the control group; high-dose group than the low-dose group of collagen type Ⅰ gene expression was significantly lower. The mRNA expression of type Ⅲ collagen in hepatic stellate cells in low and medium dose hALR groups was significantly lower than that in control group at 2, 48, 72h; the expression level of type Ⅲ collagen in hepatic stellate cells in high dose hALR group was 8, 2, 4, 48, 72h four time points were significantly lower than the control group, compared with the medium and low dose group also significantly reduced. These results suggest that recombinant human augmenter of liver regeneration can significantly inhibit the gene expression of type Ⅰ and type Ⅲ collagen in hepatic stellate cells.
