目的 研究异体耳廓移植后局部淋巴结内趋化因子MIP 3 β的含量变化及对树突状细胞的趋化作用。 方法 进行吻合血管的大鼠同种异体耳廓移植 ,ELISA法测定术后局部淋巴结MIP 3 β的含量变化趋势 ,应用改良Boyden小室进行淋巴结组织液对体外培养树突状细胞的趋化实验。结果 同系移植组术后局部淋巴结MIP 3 β含量升高 ,持续至术后第 2天 ,以后恢复至正常水平 ;异系移植组MIP 3 β含量持续上升 ,后期显著高于正常淋巴结内的表达水平 ,其对体外培养树突状细胞的趋化作用增强 ,这种趋化作用可被MIP 3 β抗体所阻断。 结论 器官移植后局部引流淋巴结内MIP 3 β表达增高 ,促进树突状细胞的迁移运动 ,可能是同种异体移植排斥反应启动与进展的重要因素
Objective To investigate the changes of chemokine MIP 3 β in local lymph nodes and the chemotactic effect on dendritic cells after allogeneic auricle transplantation. Methods The vascular anastomosis of the auricles in allogeneic rats was performed. The content of MIP 3 β in the local lymph nodes was measured by ELISA. The chemotaxis of the lymph node tissue fluid to the dendritic cells in vitro was evaluated by modified Boyden chamber. Results The content of MIP 3 β in local lymph nodes increased in the homograft group and continued to the 2nd day after the operation, and then returned to normal level. The content of MIP 3 β in the heterotopic graft group continued to increase and was significantly higher than that in the normal lymph node at the later stage , Its chemoattractant effect on dendritic cells in vitro is enhanced, and this chemotaxis can be blocked by MIP 3 β antibody. Conclusion The increased expression of MIP 3 β in local draining lymph nodes after organ transplantation may promote the migration of dendritic cells and may be an important factor in the initiation and progression of allograft rejection
