目的:观察荞麦花叶芦丁(RBFL)对糖尿病大鼠肝损害的保护作用并探讨其可能机制。方法:用高脂高热量饮食加小剂量链脲佐菌素(STZ)诱导大鼠2型糖尿病模型。设正常对照组、模型对照组、二甲双胍组(0.15g/kg)、荞麦花叶芦丁低剂量组(0.1g/kg)和荞麦花叶芦丁高剂量组(0.2g/kg),均灌胃给药,每天1次,连续6周。结果:与2型糖尿病模型组比较,RBFL高、低剂量组均可降低糖尿病大鼠的血糖值,RBFL可降低甘油三酯(TG)(P<0.05)和低密度脂蛋白胆固醇LDH-C(P<0.05),对血浆总胆固醇(TC)有降低趋势,RBFL可提高肝脏SOD活力(P<0.01),对血清SOD活力有提高的趋势。血清和肝脏MDA含量相对模型组均有显著性降低(P<0.01)。糖尿病组光镜下主要为肝细胞脂肪变性,大量胶原纤维形成。电镜下主要表现为包浆内含大量脂滴,线粒体空化,胶原纤维增生。RBFL治疗组肝组织病理改变明显好转。荞麦花叶芦丁的起效剂量为0.1g/kg,剂量范围为0.1g/kg到0.2g/kg。结论 :荞麦花叶芦丁对高脂饲料和STZ所致糖尿病大鼠肝脏损害具有一定的保护作用,其机制可能与降糖、降脂和抗氧化有关。
Objective: To observe the protective effect of buckwheat flower mosaic rutin (RBFL) on liver injury in diabetic rats and to explore its possible mechanism. Methods: Type 2 diabetic rats were induced by high fat and high calorie diet combined with low dose streptozotocin (STZ). The normal control group, model control group, metformin group (0.15g / kg), buckwheat leaf rutin low dose group (0.1g / kg) and buckwheat flower leaf rutin high dose group Stomach administration, once daily for 6 weeks. Results: Compared with model group, RBFL in high and low dose groups could reduce the blood sugar level in diabetic rats, and RBFL could decrease triglyceride (TG) (P <0.05) and low density lipoprotein cholesterol LDH-C P <0.05). The total cholesterol (TC) in plasma decreased and RBFL increased the activity of SOD in the liver (P <0.01) and the activity of SOD in the serum increased. Serum and liver MDA levels were significantly lower than the model group (P <0.01). Under light microscope in diabetic group, the majority of hepatic steatosis and collagen fibers formed. Electron microscopy mainly showed that the prolactin contained a large number of lipid droplets, mitochondria cavitation, collagen fiber hyperplasia. RBFL treatment group liver pathology improved significantly. The effective dose of buckwheat leaf rutin is 0.1 g / kg and the dosage range is 0.1 g / kg to 0.2 g / kg. CONCLUSION: Rutin in buckwheat leaves can protect liver from high fat diet and STZ - induced diabetic rats. The mechanism may be related to hypoglycemic, lipid - lowering and anti - oxidative effects.
