目的:优化乙醇对桂枝茯苓胶囊苦杏仁苷的浸出率。方法:以苦杏仁苷浸率为考核指标,单因素方差分析最佳浸出苦杏仁苷乙醇浓度。用不同浓度乙醇处理桂枝茯苓胶囊,高效液相色谱法测定各乙醇处理液中苦杏仁苷的浓度。色谱柱:(4.6mm ID×150mm,粒径5μm);流动相:甲醇和0.2%磷酸水(24∶76);检测波长:210nm;流速mL.min-1;柱温:室温;进样量15μL。结果:苦杏仁苷在0.3125~3.125μg范围内线性关系良好(r=0.9999),(n=6),60%乙醇对苦杏仁苷浸出率最高。结论:该方法重现性好,可作为桂枝茯苓胶囊乙醇浸出苦杏仁苷的简便方法。
Objective: To optimize the ethanol extraction rate of amygdalin of Guizhi Fuling capsule. Methods: Taking the rate of amygdalin as the evaluation index, single factor analysis of variance was the best method to extract the concentration of amygdalin ethanol. Guizhifuling Capsules were treated with different concentrations of ethanol, and the concentration of amygdalin in each ethanol solution was determined by high performance liquid chromatography. Column: (4.6mm ID × 150mm, particle size 5μm); mobile phase: methanol and 0.2% phosphoric acid water (24:76); detection wavelength: 210nm; flow rate mL.min-1; 15μL. Results: Amygdalin showed good linearity (r = 0.9999) in the range of 0.3125 ~ 3.125μg (n = 6), and the highest leaching rate of amygdalin was found in 60% ethanol. Conclusion: This method has good reproducibility and can be used as a simple and convenient method for extracting amygdalin from Guizhifuling capsule by ethanol.
