高胰岛素-正葡萄糖钳夹技术在比格犬中的建立及应用

目的在比格犬中建立高胰岛素-正葡萄糖钳夹技术。方法以5只健康比格犬为实验对象进行高胰岛素-正葡萄糖钳夹实验,考察血糖、胰岛素、C肽及胰高血糖素等指标。结果成功将血糖控制在目标水平,变异系数小于5%;实验过程中血清胰岛素最后稳定在(40.0±3.8)mIU/L,显著高于基础胰岛素水平;血清C肽浓度显示内源性胰岛素受到了明显的抑制,胰高血糖素没有显著升高。结论在比格犬体内建立高胰岛素-正葡萄糖钳夹技术,并将其成功用于胰岛素制剂药效学的评价。 Objective To establish a high-insulin-positive glucose clamp technique in Beagle dogs. Methods Five healthy beagle dogs were used in the experiment. The hyperinsulinemic-n-glucose clamp test was conducted to investigate the indexes of blood glucose, insulin, C-peptide and glucagon. Results The blood glucose level was successfully controlled at a target level with a coefficient of variation of less than 5%. Serum insulin levels were finally stabilized at (40.0 ± 3.8) mIU / L during the course of the experiment, significantly higher than basal insulin levels. Serum C-peptide concentrations showed that endogenous insulin was Obvious inhibition, glucagon no significant increase. Conclusion The insulin-plus-glucose clamp technique was established in Beagle dogs and successfully used in the evaluation of pharmacodynamics of insulin preparations.
