目的 观察抗震止痉胶囊联合还原性谷胱甘肽 (GSH)对帕金森病 (PD)模型大鼠旋转行为、黑质抗氧化系统及神经递质代谢的影响。方法 应用 6-OHDA立体定向注射技术制作PD大鼠模型。随机选取 2 4只模型大鼠分为 4组 (每组 4只 ) :模型组 ,中药组 ,左旋多巴 (L -dopa)组 ,中药 +GSH组 ,另设正常组 6只。分别予相应处理 ,45d后测定黑质区GSH -Px、MDA和ROS水平 ,测定尾状核DA、HVA及MAO -B水平。结果 (1)中药组和中药 +GSH组均为可部分改善PD大鼠旋转行为 (P均 <0 .0 5 ) ,二者相比无明显差异 (P >0 .0 5 )。 (2 )中药组可提高GSH -Px活性 ,降低ROS、MDA水平 (P均 <0 .0 1) ,中药 +GSH组的上述作用优于中药组 (P <0 .0 5或P <0 .0 1)。L -dopa组使GSH-Px活性降低 ,ROS和MDA水平升高 (P均 <0 .0 5 )。 (3 )中药组和中药组 +GSH组均可降低MAO -B活性 ,增加DA含量及DA/HVA比值 (P <0 .0 1或P <0 .0 0 1) ,二者相比无明显差异 (P >0 .0 5 ) ,L -dopa组MAO -B活性增强 (P <0 .0 5 ) ,DA、HVA含量及DA/HVA比值均明显升高 (P <0 .0 5或P <0 .0 1)。结论 抗震止痉胶囊联合GSH能部分改善PD模型大鼠旋转行为 ,并能减轻黑质区氧化应激损伤 ,降低MAO -B活性 ,调节纹状体DA含量及其代谢。
Objective To observe the effects of Kangzengzhijing Capsule combined with reduced glutathione (GSH) on the rotation behavior, substantia nigra antioxidant system and neurotransmitter metabolism in Parkinson disease (PD) rats. Methods PD rat model was made by 6-OHDA stereotactic injection. A total of 24 rats were randomly divided into 4 groups (4 rats in each group): model group, traditional Chinese medicine group, L-dopa group, TCM + GSH group, and another 6 normal rats. After 45 days, the levels of GSH-PX, MDA and ROS in substantia nigra were measured, and the levels of DA, HVA and MAO-B in caudate nucleus were measured. Results (1) Both TCM and TCM + GSH groups could partially improve the rotational behavior of PD rats (all P <0.05), there was no significant difference between the two groups (P> 0.05). (2) The activity of GSH-Px and the level of ROS and MDA were all decreased in the traditional Chinese medicine group (P <0.01 or P <0.05). 0 1). L-dopa group decreased GSH-Px activity and ROS and MDA levels (all P <0.05). (3) The activity of MAO-B, the content of DA and the ratio of DA / HVA were both decreased in the traditional Chinese medicine group and the traditional Chinese medicine group + GSH group (P <0.01 or P <0.01) (P <0.05), the activity of MAO-B in L-dopa group was significantly increased (P <0.05), the contents of DA, HVA and DA / HVA were significantly increased (P <0.05 or P <0 .0 1). Conclusion Seizurezhijing capsules combined with GSH can partly improve the rotation behavior of rats with PD and reduce the oxidative stress injury in substantia nigra and decrease the activity of MAO-B and the DA content and metabolism in striatum.
