目的研究小鼠隐睾模型中睾丸的质量及组织病理学变化,并探讨其临床意义。方法取50只6周龄ICR雄性健康小鼠,随机选取25只,造左侧隐睾模型,视为隐睾组;另外25只小鼠,左侧睾丸适度牵拉仍保持原位置不动,视为假手术组。再随机分成5组,每组包含5只隐睾小鼠,5只假手术小鼠。术后3d、7d、14d、21d和28d分别颈椎离断处死各组小鼠,记录每只小鼠的体质量和两只睾丸的质量,观察睾丸的组织病理学变化并依据Johnsen评分标准进行评分。结果(1)各个时间段隐睾组小鼠与假手术组体质量的比较无统计学意义(P>0.05):(2)隐睾组小鼠两侧睾丸脏体比(脏器质量/体质量×1 000)的比较均有统计学意义,假手术组两侧睾丸脏体比的比较无统计学意义(P>0.05);(3)隐睾的质量从第7天开始减少并以一种时间依赖方式递减,差异有统计学意义(P<0.05)。与此同时,假手术组两侧睾丸质量改变不具有统计学意义(P>0.05);(4)Johnsen评分从术后第3天开始,隐睾侧睾丸的评分均低于其对侧睾丸且呈递减趋势;(5)睾丸的组织病理学变化与睾丸的质量变化一致,第7天生精细胞开始凋亡并以一种时间依赖的方式凋亡。结论实验性隐睾小鼠模型表明隐睾在腹腔中以一种时间依赖方式在改变,存在一个时间段,可能是实施干预治疗最恰当的时机,有望更好的恢复睾丸的生精功能。
Objective To study the testis quality and histopathological changes in mouse cryptorchidism model and to explore its clinical significance. Methods Fifty healthy 6-week-old ICR male mice were randomly selected and 25 rats were treated with left cryptorchidism model as cryptorchidism group. In the other 25 mice, the left testes maintained their original positions with moderate traction, Regarded as sham operation group. Then randomly divided into 5 groups, each group contains 5 cryptorchid mice, 5 sham mice. At 3, 7, 14, 21, and 28 days after operation, mice in each group were sacrificed by cervical spine disconnection. The body weight and the weight of two testis were recorded. The histopathological changes of testis were observed and scored according to Johnsen score . Results (1) There was no significant difference in body weight between cryptorchid group mice and sham operation groups at each time point (P> 0.05) :( 2) (P> 0.05). (3) The quality of cryptorchidism decreased from the 7th day to the end of the 7th day Time-dependent manner decreased, the difference was statistically significant (P <0.05). At the same time, testicular mass changes on both sides of the sham-operated group were not statistically significant (P> 0.05). (4) The scores of Johnsen score on the testis side of cryptorchidism were lower than those of the contralateral testis (5) The histopathological changes of testis were consistent with the changes of testis quality. On day 7, spermatogenic cells began to apoptosis and apoptosis occurred in a time-dependent manner. Conclusion The experimental cryptorchid mouse model shows that cryptorchidism changes in a time-dependent manner in the peritoneal cavity for a period of time, which may be the most appropriate time for interventional therapy. It is expected to restore the function of testis spermatogenesis better.
