目的:建立保健食品中金丝桃苷测定的高效液相色谱(HPLC)方法。方法:试验采用迪马钻石Ⅱ(4.6 mm×250 mm,5μm)色谱柱,乙腈-0.2%磷酸水溶液为流动相洗脱,在波长为256 nm下进行检测。结果:方法的线性范围:9.37 g/ml~187 g/ml。相关系数r为0.9999;检出限:0.4μg/g;定量限:1.2μg/g。高低两个浓度水平加标回收率:87%~94%;相对标准偏差(RSD)1.8%。结论:本方法简便、快速、准确、重现性好,适用于保健食品中金丝桃苷的测定。
Objective: To establish a high performance liquid chromatography (HPLC) method for the determination of hyperin in health food. Methods: Dima Diamond Ⅱ (4.6 mm × 250 mm, 5 μm) column and acetonitrile - 0.2% phosphoric acid aqueous solution were used as the mobile phase to elute at a wavelength of 256 nm. Results: The linear range of the method was 9.37 g / ml to 187 g / ml. The correlation coefficient r was 0.9999; the detection limit was 0.4μg / g; the limit of quantification was 1.2μg / g. High and low levels of the two spiked recoveries: 87% to 94%; relative standard deviation (RSD) 1.8%. Conclusion: The method is simple, rapid, accurate and reproducible. It is suitable for the determination of hyperoside in health food.
