目的 环孢素亲合素 (cyclophilin ,CyP)是细胞内环孢霉素A(CsA)的受体 ,探讨CyP在肾病综合征患儿中基因表达及临床意义。方法 采用逆转录聚合酶链反应 (RT PCR)测定 2 8例肾病综合征患儿血白细胞中CyPmRNA的表达 ,并以正常儿童作对照。 结果 CyP碱基对 4 0 8bp与 β Actin条带 2 3 4bp可清晰区分。不同类型肾病综合征患者血白细胞中CyPmRNA表达的水平无差异 ,同时发现肾病综合征患儿急性期血白细胞中CyP基因表达水平明显高于恢复期与对照组。 结论 肾病综合征患儿急性期有高水平的CyPmRNA表达 ,在此时给予合适剂量环孢素亲合素的反应物(如CsA治疗 )可促进肾病综合征的缓解。
Objective Cyclophilin (cyclophilin, CyP) is an intracellular cyclosporine A (CsA) receptor, CyP in children with nephrotic syndrome gene expression and clinical significance. Methods The expression of CyP mRNA in blood leukocytes of 28 children with nephrotic syndrome was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and compared with normal children. Results CyP base pair 408 bp and β Actin band 233 bp can be clearly distinguished. The levels of CyP mRNA in leukocytes of patients with different types of nephrotic syndrome were no difference. The levels of CyP gene in acute leukopenia patients with nephrotic syndrome were significantly higher than those in convalescent and control groups. Conclusions Children with nephrotic syndrome have a high level of CyP mRNA expression in acute stage. At this time, the appropriate dose of cyclosporin A (such as CsA) can promote nephrotic syndrome.
