目的 在体外研究白色念珠菌临床分离株对人肺上皮细胞的黏附作用及其影响因素。方法 将不同浓度的白色念珠菌和人肺上皮细胞在体外作用不同的时间 ,固定后经革兰染色 ,在倒置显微镜下计数白色念珠菌黏附人肺上皮细胞的黏附数。观察胰酶、白色念珠菌培养上清液、加热等处理因素对白色念珠菌黏附人肺上皮细胞的影响。结果 白色念珠菌的黏附与浓度、时间有关 ,在 5× 10 3~ 5× 10 7CFU/ml范围内 ,黏附数随浓度的增加而增加。该黏附作用 15min后趋于饱和。在 15min内 ,白色念珠菌对肺上皮细胞的黏附随时间的延长而增强。白色念珠菌经胰酶或 10 0℃ 30min处理后黏附数降低 ,培养上清液及上清液经加热或胰酶处理后都能抑制白色念珠菌的黏附作用。结论 白色念珠菌对人肺上皮细胞的黏附作用与浓度和时间有依赖的关系 ,是特异性黏附过程。不同因素处理白色念珠菌和肺上皮细胞 ,其黏附数发生改变 ,说明黏附作用与白色念珠菌表面黏附因子和人肺上皮细胞表面的黏附受体的数量和结构有关。
Objective To study the adhesion of Candida albicans clinical isolates to human lung epithelial cells in vitro and its influencing factors. Methods Different concentrations of Candida albicans and human lung epithelial cells were exposed to different times in vitro. Gram stain was performed after fixation. The number of Candida albicans adherent human lung epithelial cells was counted under an inverted microscope. To observe the influence of trypsin, Candida albicans culture supernatant, heating and other treatment factors on Candida albicans adhesion to human lung epithelial cells. Results The adhesion of C. albicans was related to the concentration and time. The number of adhesion increased with the increase of concentration in the range of 5 × 10 3 ~ 5 × 10 7 CFU / ml. The adhesion tended to be saturated after 15 min. Within 15 min, C. albicans adhesion to the lung epithelial cells increased with time. Candida albicans after trypsin or 10 0 ℃ 30min treatment reduced the number of adhesion, culture supernatant and supernatant after heating or trypsin treatment can inhibit the adhesion of Candida albicans. Conclusion Candida albicans adhesion to human lung epithelial cells in a concentration-dependent and time-dependent manner is a specific adhesion process. Different factors in the treatment of Candida albicans and lung epithelial cells, the number of adhesion changes, indicating that the adhesion of Candida albicans surface adhesion factor and human lung epithelial cell adhesion receptors on the number and structure.
