目的评价通心络对高血压、高血糖合并高胆固醇血症大鼠模型动脉功能的影响及机制。方法将体重300~400 g的成年自发性高血压大鼠(SHR)32只随机分为普食组(SHR组)、对照组(模型组)、低剂量通心络(0.2 g/kg)干预组(通心络低组)、高剂量通心络(0.8 g/kg)干预组(通心络高组),另取8只正常血压大鼠作为正常组,高脂喂养4周建立动物模型。通心络干预4周后,行主动脉离体血管功能检查,用荧光定量PCR方法测定过氧化物酶体增殖物受体γ(PPAR-γ)mRNA表达。结果通心络干预后大鼠离体主动脉对乙酰胆碱、硝普钠引起的舒张反应增强,通心络高组对A23187的舒张反应增强(P<0.05),通心络剂量依赖性增加PPAR-γ基因表达。结论通心络干预具有保护血管内皮功能及逆转血管平滑肌功能障碍等作用,其机制可能与激活PPAR-γ基因表达有关。
Objective To evaluate the effect and mechanism of Tongxinluo on arterial function in hypertensive and hyperglycemic rats with hypercholesterolemia. Methods Thirty-two adult spontaneously hypertensive rats (SHR) weighing 300-400 g were randomly divided into normal group (SHR group), control group (model group), low dose Tongxinluo (0.2 g / kg) (Tongxinluo low group), high dose Tongxinluo (0.8 g / kg) intervention group (Tongxinluo high group), another 8 normal rats as normal group and high fat diet for 4 weeks to establish animal models . Tongxinluo intervention four weeks later, the aortic ex vivo vascular function tests, using fluorescent quantitative PCR method to determine peroxisome proliferator receptor gamma (PPAR-gamma) mRNA expression. Results After the intervention of Tongxinluo, the relaxation responses of isolated aorta to acetylcholine and sodium nitroprusside in rats increased, the relaxation of A23187 in Tongxinluo high group was enhanced (P <0.05), Tongxinluo increased PPAR- γ gene expression. Conclusion Tongxinluo intervention can protect vascular endothelial function and reverse vascular smooth muscle dysfunction and its mechanism may be related to the activation of PPAR-γ gene expression.
