采用GPY培养基对格木内生真菌烟曲霉Aspergillus fumigatus菌株进行培养发酵,发酵产物经冻融处理后分离得到菌液与菌丝体。菌丝体经乙醇提取后,分别用乙酸乙酯和正丁醇萃取,根据肿瘤细胞毒活性筛选结果,选择菌体的乙酸乙酯萃取物进行化学成分研究。运用硅胶色谱柱、凝胶Sephadex LH-20柱色谱、反相柱色谱和制备HPLC等多种色谱技术,采用波谱学方法从中分离鉴定了5个二酮哌嗪化合物,其结构分别为cyclo-(R-Pro-R-Phe)(1),cyclo-(trans-4-OH-D-Pro-D-Phe)(2),cyclo-(R-Tyr-S-Ile)(3),cyclo-(R-Phe-S-Ile)(4),cyclo-(R-Val-S-Tyr)(5)。
The GPY culture medium was used to culture and ferment Aspergillus fumigatus, an endophytic fungus of Pleurotus ostreatus. The fermentation products were separated by freeze-thaw and the mycelium and mycelium were isolated. After the mycelium was extracted by ethanol, the mycelia were extracted with ethyl acetate and n-butanol, respectively. According to the results of screening cytotoxic activity of the tumor cells, the ethyl acetate extracts of the bacterial cells were selected for chemical constituents study. Five diketopiperazine compounds were isolated and identified by spectroscopic methods using silica gel column, gel Sephadex LH-20 column chromatography, reverse phase column chromatography and preparative HPLC. Their structures were cyclo- ( Cyclo- (trans-4-OH-D-Pro-D-Phe) (2), cyclo- (R-Tyr-S-Ile) (R-Phe-S-Ile) (4), cyclo- (R-Val-S-Tyr) (5).
