本研究观察了α-黑色素细胞刺激素(α-MSH)对家兔白细胞介素-1β(IL-1β)发热效应及下丘脑组织腺苷环-磷酸(cAMP)含量的影响;同时观察了下丘脑组织体外培养过程中,α-MSH对IL-1β刺激下丘脑释放cAMP的影响。结果显示:α-MSH能显著降低IL-1β引起的体温升高(P<0.05);同时抑制下丘脑组织cAMP含量的增高(P<0.01)。IL-1β与下丘脑组织培养,其上清液中cAMP含量明显升高(P<0.01);而预先加入α-MSH,再加同量IL-1β培养,则培养上清液的cAMP含量接近对照组。体内外实验表明,α-MSH抑制中枢发热介质cAMP,的生成可能是其解热的重要机制之一.
In this study, we observed the effect of α-MSH on the fever and the content of adenosine ring-and-cAMP in hypothalamus of rabbits. Effect of α-MSH on cAMP released from hypothalamus stimulated by IL-1β in cultured thalamus tissue. The results showed that α-MSH could significantly decrease the body temperature induced by IL-1β (P <0.05) and inhibit the increase of cAMP in hypothalamus (P <0.01). IL-1β and hypothalamus tissue culture, the supernatant cAMP content was significantly increased (P <0.01); pre-addition of α-MSH, plus the same amount of IL-1β culture, the culture supernatant cAMP Content close to the control group. In vitro and in vivo experiments show that α-MSH inhibition of the central heating medium cAMP, the generation may be one of its important mechanisms of antipyretic.
