目的:对加味小柴胡汤颗粒及其有效物质部位组方进行抗胃溃疡的药效研究,为其临床疗效提供科学依据,并为开发加味小柴胡汤颗粒医院制剂奠定基础。方法:构建大鼠幽门结扎型、醋酸刺激型及小鼠无水乙醇诱发胃溃疡等动物模型,以加味小柴胡汤颗粒及其有效物质部位组方作为供试药物进行试验研究。观察其对模型动物的治疗作用,以及对胃液分泌、胃酸含量、胃蛋白酶活性、超氧化物歧化酶活性(SOD)和丙二醛(MDA)含量的影响。结果:加味小柴胡汤颗粒及其有效部位组方,与空白模型组相比,均能明显降低胃溃疡指数(P<0.01)、胃液总酸度(P<0.01)和胃蛋白酶活性(P<0.05或P<0.01),提高SOD活力(P<0.05),抑制MDA升高(P<0.05)。结论:加味小柴胡汤颗粒及其有效部位组方对急、慢性胃溃疡均具有较好的药效作用,其作用机制与降低胃液总酸度及胃蛋白酶活性、提高SOD活力,抑制MDA升高等有关。
OBJECTIVE: To study the anti-gastric ulcer efficacy of Modified Chaihu Hu Tang Granules and its active substance sites to provide a scientific basis for its clinical curative effect and lay a foundation for the development of the formulation of Modified Xiao Chai Hu Tang Granules Hospital. Methods: Animal models of gastric ulcer caused by pylorus ligation in rats, acetic acid stimulation and mice ethanol were constructed. The experimental group was treated with Jiawei Xiao Chai Hu Tang Granules and its effective substances. To observe its therapeutic effect on model animals, as well as gastric juice secretion, gastric acid content, pepsin activity, superoxide dismutase activity (SOD) and malondialdehyde (MDA) content. Results: Compared with the blank model group, the modified Xiaochaihu decoction granules and the effective fractions thereof all significantly reduced the gastric ulcer index (P <0.01), the gastric acid total acidity (P <0.01) and the pepsin activity (P <0.05) Or P <0.01), increased SOD activity (P <0.05) and inhibited MDA (P <0.05). Conclusion: Modified Xiaochaihu decoction and its effective fractions have good effect on both acute and chronic gastric ulcer. Its mechanism of action is related to reducing the total acidity and pepsin activity of gastric juice, increasing the activity of SOD and inhibiting the increase of MDA .
