目的:探讨干扰素-γ不同剂量、不同作用时间对小鼠白血病细胞FBL-3体外侵袭能力的影响。方法:小鼠白血病细胞株FBL-3分为6组,其中A组为空白对照组;B~E组分别用干扰素-γ5,50,200,500 ng/mL处理48 h;F组为干扰素-γ50 ng/mL处理2周。应用Transwell侵袭模型检测各组FBL-3细胞的体外侵袭力,采用单因素方差分析进行比较。结果:与A组比较,C、D和E组穿过Matrigel胶的FBL-3细胞数目明显减少,且存在剂量依赖性效应(P<0.05)。F组细胞侵袭能力增强,明显高于对照组(P<0.05)。结论:干扰素-γ短期作用(48 h)可以剂量依赖性方式明显抑制FBL-3细胞的体外侵袭能力;小剂量干扰素-γ长期作用可促进FBL-3细胞的侵袭能力。
Objective: To investigate the different doses of interferon-γ, different time on the impact of mouse leukemia cells FBL-3 invasion in vitro. Methods: FBL-3 cells were divided into 6 groups: group A was blank control group; group B was treated with interferon-γ5,50,200,500 ng / mL for 48 h; group F was interferon-γ50 ng / mL for 2 weeks. The invasiveness of FBL-3 cells in each group was detected by Transwell invasion model and compared by one-way ANOVA. Results: Compared with group A, the number of FBL-3 cells in Matrigel group decreased obviously in groups C, D and E, and there was a dose-dependent effect (P <0.05). F cell invasion ability was significantly higher than that of the control group (P <0.05). CONCLUSION: The short-term effect of IFN-γ (48 h) can significantly inhibit the in vitro invasiveness of FBL-3 cells in a dose-dependent manner. The long-term effects of IFN-γ can promote the invasion ability of FBL-3 cells.
