目的:研究基因BMI1在中枢神经系统最常见的肿瘤-胶质瘤组织中的表达情况,并探讨其表达水平与胶质瘤的恶性程度及发生、发展的关系。方法:使用荧光定量实时PCR技术检测基因BMI1在34例各级别胶质瘤手术切除组织及10例正常对照组织中的表达。同时对34例各级别胶质瘤组织及10例正常对照脑组织使用免疫组化的方法检测BMI1蛋白的表达。结果:实时PCR的结果显示,基因BMI1在对照脑组织、Ⅱ级胶质瘤、Ⅲ级胶质瘤及Ⅳ,级胶质瘤组织中的表达相对值分别为0.655、2.65、5.62及5.43。胶质瘤组中BMI1的表达水平显著高于对照组(P=0.001).恶性胶质瘤组(Ⅲ级与Ⅳ级胶质瘤)中BMI1的表达水平显著高于良性胶质瘤(Ⅱ级胶质瘤),JP=0.015,Ⅲ级胶质瘤组与Ⅳ级胶质瘤组之间未发现存在差异(P=0.902);免疫组化结果显示,BMI1蛋白在对照脑组织、Ⅱ级胶质瘤、Ⅲ级胶质瘤和Ⅳ级胶质瘤组织中的表达阳性率分别为10%、25%、100%和100%。各级别胶质瘤组织中BMI1蛋白的阳性表达率显著高于正常脑组织对照组(P=0.001),而且在恶性胶质瘤组织中BMI1蛋白的阳性表达率显著高于良性胶质瘤(P=0.000)。结论:无论是在mRNA水平还是在蛋白水平上,BMI1在各级别脑胶质瘤标本中的表达水平均明显高于正常脑组织对照,且恶性胶质瘤中BMI1的表达要明显高于良性胶质瘤。BMI1在胶质瘤的发生、发展过程中有重要作用。
OBJECTIVE: To study the expression of BMI1 gene in the most common tumor of glioma tissues of the central nervous system and to explore the relationship between the expression of BMI1 gene and the malignant degree, occurrence and development of gliomas. Methods: Fluorescent quantitative real-time PCR was used to detect the expression of BMI1 in 34 cases of glioma and 10 cases of normal control tissues.
