新型可降解镁锌合金材料应用于动静脉瘘封堵材料的动物研究

目的 构建犬动静脉瘘动物模型,对新型可降解镁锌合金材料植入动静脉瘘动物模型后生物相容性及安全性进行研究.方法 实验用比格犬10只,构建犬髂内动静脉瘘动物模型,分为实验组(镁锌合金材料植入动静脉瘘)5只和对照组(仅动静脉瘘造模)5只,比较两组实验动物饮食、活动及切口反应情况,分别于术前及术后1、4、8及12周检测血清肝肾功电解质(肌酐、谷丙转氨酶、总胆红素、尿素氮及血镁锌浓度)生化指标,按照计划在术后12周处死实验犬,取出动静脉瘘标本及肺肝肾心等重要脏器进行苏木精-伊红(HE)染色.结果 两组实验动物饮食、活动正常、切口未见明显感染迹象.术前及术后谷丙转氨酶、总胆红素及尿素氮浓度差异无统计学意义(P>0.05);血镁浓度在术后1周、血锌及肌酐浓度在术后1、4周差异有统计学意义(P0.05).术后12周 HE 染色示实验组动静脉瘘处动脉内膜较对照组稍增生,未发现明显炎症细胞浸润;肾小管和间质未出现异常形态、肿胀及明显炎症反应;肺泡结构和功能完整,未发现明显血栓形成;肝、心细胞未出现水肿及坏死.结论 新型可降解镁锌合金材料植入犬动静脉模型后,对血液生化指标及重要脏器结构功能无明显影响,可作为动静脉瘘的理想封堵材料,具有良好的前景及临床应用价值.“,”Objective To evaluate the biocompatibility and safety of the new biodegradable Mg-Zn alloy used as occluder materials in dogs with aortic-vena cava fistula. Methods Ten beagle dogs were used to establish aortic-vena cava fistula model. They were equally divided into experimental group (Mg-Zn alloy implanted aortic-vena cava fistula)(n=5)and control group(noting implanted aortic-vena cava fistula)(n=5). The diet, activity and incision response of dogs were observed. Pre-and post-operationally, the blood biochemical test, serum magnesium and zinc were compared between the two groups. After 12 weeks, all beagles were executed by lethal dose of pentobarbital after removal of the heart, liver, lung, kidney and roughly 5 cm of aortic-vena cava fistula specimen adopting by hematoxylin and eosin(HE) staining. Results The diet and activity of experimental animals between two groups were normal, and inci-sion was not obvious signs of infection. Pre-and post-operationally, indicators such as ALT、TBiL and BUN were found no statistically significant differences in the experimental group compared to the control group at each time point(P>0.05). Although serum magnesium values in the experimental group in the first week were gently higher than the control group(P0.05). The serum zinc Scr levels in the experimental group were higher than the control group in the first and fourth week respectively(P0.05). Though HE staining assessment, all aortic-vena cava fistula specimens manifested no significant inflammatory response with slightly thickened intima, without obvious inflammatory cell infiltration. The tubules and interstitium of kidney did not present abnormal morphology, without swelling or inflammatory re-action. The structure and function of alveoli was complete and no PE was observed. Hepatocytes did not show obvious edema or necrosis. Apoptosis or necrosis of myocardial cell was not detected, neither collagen proliferation nor inflammatory cell infiltration was detected. Conclusion New biodegradable Mg-Zn alloy does not harm the function of important organs and blood biochemical indicators. It may be a perfect candi-date of biodegradable and safe material of plugging device.
