目的:目的:探讨黄酮类化合物槲皮素对人鼻咽癌HEN1细胞系的抑制增殖和诱导凋亡的作用,并对其机制做初步探讨。方法:应用MTT法检测不同浓度槲皮素对人鼻咽癌HEN1细胞的增殖抑制作用;应用流式细胞仪检测细胞凋亡率及细胞周期分布;比色法测定凋亡相关蛋白caspase-3的表达。结果:槲皮素能明显抑制人鼻咽癌HEN1细胞增殖,存在剂量依赖(r=0.709;P<0.01)与时间依赖(r=0.703;P<0.01);HEN1细胞经不同浓度槲皮素(15、30、60、90、120μmol/L)作用48 h后细胞凋亡率分别为4.81%、9.02%、14.33%、19.65%、28.88%,明显高于对照组(1.70%)(P<0.01);随着槲皮素浓度升高,凋亡细胞和坏死细胞比例增加,将细胞特异性地阻滞在G2/M期,出现凋亡峰;槲皮素组的HEN1细胞凋亡相关因子caspase-3表达明显高于对照组(P<0.05)。结论:槲皮素能通过caspase-3途径抑制人鼻咽癌HEN1细胞增殖,诱导细胞凋亡,并具有细胞周期特异性。
OBJECTIVE: To investigate the effect of quercetin, a flavonoid, on the proliferation and apoptosis induction of human nasopharyngeal carcinoma HEN1 cell line, and to explore its mechanism. METHODS: MTT assay was used to detect the inhibitory effect of different concentrations of quercetin on the proliferation of human nasopharyngeal carcinoma HEN1 cells. Flow cytometry was used to detect the apoptosis rate and cell cycle distribution. The colorimetric method was used to determine the apoptosis-related protein caspase-3. expression. RESULTS: Quercetin significantly inhibited the proliferation of human nasopharyngeal carcinoma HEN1 cells in a dose-dependent manner (r=0.709; P<0.01) and time-dependent (r=0.703; P<0.01); HEN1 cells were treated with different concentrations of quercetin ( The apoptosis rates of cells treated with 15, 30, 60, 90, and 120 μmol/L for 48 h were 4.81%, 9.02%, 14.33%, 19.65%, and 28.88%, respectively, which were significantly higher than those in the control group (1.70%) (P<0.01).
