目的评价反义LeETR1转基因番茄(ETR1)向动物肠道菌群和子代动物转移外源基因的能力及对小鼠生殖发育的影响。方法SD大鼠和ICR小鼠连续饲喂ETR1、非转基因番茄(B1)和蒸馏水30 d后,分别进行大鼠盲肠菌群培养物的抗生素耐药性试验及小鼠生殖毒性试验,提取耐药阳性菌、亲代和子代小鼠的基因组进行外源基因特异性聚合酶链式反应法(PCR)。结果肠道菌的耐药性和小鼠生殖发育情况无明显改变,在耐药菌和两代小鼠体内均未发现由受试物引入的外源基因。结论ETR1对小鼠无生殖毒性,其携带的外源基因不能向动物体细胞、肠道菌及子代动物转移。
Objective To evaluate the ability of antisense LeETR1 transgenic tomato (ETR1) to transfer exogenous genes into gut flora and progeny of animals and its effect on reproductive development in mice. Methods SD rats and ICR mice were fed with ETR1, non-transgenic tomato (B1) and distilled water for 30 days, respectively, antibiotic resistance test and reproductive toxicity test of cecal flora in rats were carried out to extract drug-resistant Positive bacteria, parental and offspring mice were subjected to exogenous gene-specific polymerase chain reaction (PCR). Results The resistance of enteric bacteria and reproductive development in mice showed no significant changes, and no foreign genes introduced by the test substance were found in both drug-resistant bacteria and two generations of mice. Conclusion ETR1 has no reproductive toxicity to mice and the exogenous gene it carries can not transfer to somatic cells, intestinal bacteria and progeny of animals.
