目的:探讨胃硬癌中Wnt/β-catenin信号通路的活化及与间质纤维化的关系。方法:采用免疫组化染色SP方法,检测29例胃硬癌(胃硬癌组)、29例胃非硬癌(非硬癌组)及30例癌旁良性胃组织(对照组)中Wnt2及β-catenin的表达,应用苦味酸-天狼猩红染色方法检测Ⅰ型与Ⅲ型胶原纤维在以上三组中的表达。结果:Wnt2在胃硬癌和非硬癌组织中表达水平明显高于对照组(P<0.01)。胃硬癌组中β-catenin在细胞核中的表达明显高于非硬癌组及对照组,三组间均存在显著性差异(P<0.05)。Ⅰ型与Ⅲ型胶原的表达与β-catenin胞膜的表达呈负相关(P<0.01),与β-catenin胞质胞核的表达均呈正相关(P<0.01)。结论:Wnt/β-catenin信号通路的活化参与胃癌的发生发展,并可能参与调节胃硬癌间质纤维化的发生过程。
Objective: To investigate the activation of Wnt / β-catenin signaling pathway and its relationship with interstitial fibrosis in gastric sclerosis. Methods: Immunohistochemical SP method was used to detect the expression of Wnt2 and its mRNA in 29 patients with gastric cancer (gastric cancer), 29 patients with non-gastric cancer (non-cancer) and 30 patients with adjacent normal tissues β-catenin expression, picric acid-Sirius red staining method to detect type Ⅰ and type Ⅲ collagen fibers in the above three groups. Results: The expression of Wnt2 in gastric cancer and non-cancer tissues was significantly higher than that in the control group (P <0.01). The expression of β-catenin in the gastric cancer group was significantly higher than that in the non-cancer group and the control group, and there was significant difference among the three groups (P <0.05). The expressions of type Ⅰ and type Ⅲ collagens were negatively correlated with the expression of β-catenin (P <0.01), and positively correlated with the expression of β-catenin in cytoplasmic nuclei (P <0.01). Conclusion: The activation of Wnt / β-catenin signaling pathway is involved in the development of gastric cancer and may be involved in the regulation of the development of interstitial fibrosis in gastric cancer.
