全合成甘露糖化壳聚糖,为靶向递送药物至巨噬细胞和树突状细胞提供载体材料。实验以甘露糖为起始原料,通过6步反应合成预期取代度为5%~10%的甘露糖化壳聚糖,分别用红外(FTIR)、元素分析等技术对其结构进行确证和表征,并通过HeLa和A549细胞毒性试验评价其细胞毒性。结果表明,所制备的甘露糖化壳聚糖为橘黄色固体,不溶于水,在pH低于6.0的醋酸盐缓冲液中溶解良好,取代度为7.35%;其细胞毒性较低,当浓度达到100μg/ml时,细胞存活率大于80%。合成的甘露糖化壳聚糖是一种安全的靶向递送载体。
Fully synthetic mannosylated chitosan provides a carrier material for targeted delivery of drugs to macrophages and dendritic cells. In the experiment, mannose was used as the starting material to synthesize mannosylated chitosan with the expected substitution degree of 5% ~ 10% by 6 steps. The structure of the product was confirmed and characterized by FTIR and elemental analysis respectively. Cytotoxicity was assessed by HeLa and A549 cytotoxicity assays. The results showed that the mannosylated chitosan prepared was an orange solid and insoluble in water. The mannosyl chitosan prepared was well dissolved in acetate buffer at a pH of less than 6.0 with a degree of substitution of 7.35%. The cytotoxicity was low. When the concentration reached At 100 μg / ml, cell viability was greater than 80%. Synthetic mannosylated chitosan is a safe targeting delivery vehicle.
