目的观察氯化镉(CdCl2)处理人肝癌SMMC-7721细胞株某些生化指标的变化。方法利用四甲基偶氮唑蓝(MTT)法检测细胞存活率;利用生化法检测CdCl2处理人肝癌SMMC-7721细胞株丙二醛(MDA)含量和超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-px)活力。结果不同浓度的氯化镉作用12、24和48 h,随着给予CdCl2剂量的增加,细胞存活率显著下降。随着给药时间的延长,对细胞抑制作用增强,存在着剂量-效应关系、时间-效应关系;给予细胞10~40μmol/l CdCl2作用24 h,随着给镉剂量增加,细胞的MDA含量增加,SOD和GSH-px的活力下降。结论CdCl2可致人肝癌SMMC-7721细胞株存活率下降和某些氧化和抗氧化生化指标改变有关。
Objective To observe the changes of some biochemical indexes of human hepatoma SMMC-7721 cells treated with cadmium chloride (CdCl2). Methods The cell viability was detected by MTT assay. The contents of malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione Glycopeptide peroxidase (GSH-px) activity. Results After cadmium chloride treatment at different concentrations for 12, 24 and 48 h, cell viability decreased significantly with the increase of CdCl2 dose. With the prolongation of administration time, the inhibitory effect on cells increased, there was a dose-response relationship, time-effect relationship; given cells with 10 ~ 40μmol / l CdCl2 for 24 h, with increasing cadmium dose, the cell MDA content increased , The activity of SOD and GSH-px decreased. Conclusion CdCl2 can reduce the survival rate of human hepatocellular carcinoma SMMC-7721 cells and some changes of some oxidative and anti-oxidative biochemical indexes.
