目的成功建立脾气虚变应性鼻炎(AR)大鼠模型,应用温阳益气方干预,探讨温阳益气方治疗脾气虚AR的作用机理,为临床提供实验依据。方法利用大黄泻下法等综合因素加上卵清蛋白建立脾气虚AR动物模型,应用温阳益气方进行药物干预,观察实验大鼠鼻部症状、检测鼻分泌物及血嗜酸性粒细胞(EOS)、血清IL-5、嗜酸性细胞阳离子蛋白(ECP)含量。结果大鼠模型的鼻部症状(鼻痒、喷嚏、清涕)、鼻分泌物及血EOS、血清IL-5、ECP含量升高(P<0.01);而经过温阳益气方干预后的脾气虚AR大鼠组症状减轻,血清IL-5、ECP含量降低(P<0.01)。结论温阳益气方能降低脾气虚AR大鼠模型血清IL-5和ECP水平,防止EOS活化、增殖、分化,并防止EOS释放ECP,对治疗AR有作用。
Objective To establish a rat model of spleen-qi-deficiency allergic rhinitis (AR) successfully and to explore the mechanism of warming yang qi recipe in treating spleen-qi-deficiency syndrome (AR) by means of the intervention of warming yang qi and providing the experimental evidence for clinical practice. Methods The animal model of asthenia induced by spleen deficiency was established by the combination of rhubarb and other methods with ovalbumin. The rats were anesthetized with warming yang qi for drug intervention. Nasal symptoms, nasal secretions and blood eosinophils (EOS) , Serum IL-5, eosinophil cationic protein (ECP) content. Results Nasal symptoms (nasal itching, sneezing, clear nasal discharge), nasal secretions and blood EOS, serum IL-5 and ECP contents increased in the rat model (P <0.01) Symptoms of AR rats were relieved, serum IL-5 and ECP levels were decreased (P <0.01). CONCLUSION WenYiNiQi can reduce the level of IL-5 and ECP in serum of rats with spleen-qi-deficiency syndrome and prevent the activation, proliferation and differentiation of EOS, and prevent EOS from releasing ECP.
