无患子皂苷对大鼠心肌缺血的保护作用

目的:观察无患子皂苷对大鼠实验性心肌缺血损伤的保护作用。方法:采用结扎冠脉左前降支的方法复制大鼠心肌缺血模型,将造模成功的大鼠随机均分为模型组、阳性对照组(三七总皂苷0.081 g·kg-1)、无患子皂苷低、中、高剂量组(0.054,0.162,0.486 g·kg-1),每组10只,另取假手术组10只,连续ig给药28 d。分别监测各组大鼠手术前及手术后即刻,1,3,7,10,14,21,28 d的心电图,并测定给药28 d后的心肌梗死率及血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)的含量。结果:无患子皂苷各剂量组大鼠心电图S-T段的抬高程度均显著下降,心肌梗死范围明显缩小,血清SOD和GSH-Px的活性显著增强,MDA含量显著降低。结论:无患子皂苷能有效保护大鼠实验性心肌缺血损伤,其作用可能与清除氧自由基,提高抗氧化酶活性及抗脂质过氧化有关。 Objective: To observe the protective effect of Sapindus saponins on experimental myocardial ischemia in rats. Methods: The rat model of myocardial ischemia was established by ligating the left anterior descending coronary artery. The successful model rats were randomly divided into model group, positive control group (Panax Notoginseng Saponins 0.081 g · kg -1), Mice in low, medium and high doses of Saponins (0.054,0.162,0.486 g · kg-1), 10 in each group, and another 10 sham-operated group were given ig for 28 days. The electrocardiogram of rats in each group were monitored before operation and immediately after operation. The electrocardiogram was measured on the 1st, 3rd, 7th, 10th, 14th, 21st and 28th day after the operation. The myocardial infarction rate and the level of superoxide dismutase (SOD ), Glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) content. Results: The S-T elevation of electrocardiogram was significantly decreased in each dose group of Sapindus saponin, the range of myocardial infarction was significantly reduced, the activities of serum SOD and GSH-Px were significantly increased, and the content of MDA was significantly decreased. Conclusion: Sapindus saponin can effectively protect experimental myocardial ischemia injury in rats, which may be related to scavenging oxygen free radicals, increasing antioxidant enzyme activity and anti-lipid peroxidation.
