目的探讨雪灵芝粗多糖(arenaria kansuensis crude polysaccharide,AKCP)对环磷酰胺致免疫抑制小鼠免疫功能的影响。方法将小鼠随机分为5组:空白对照组、环磷酰胺(CTX)模型组、AKCP高、中、低剂量组。除空白对照组外其他4组采用腹腔注射CTX建立小鼠免疫低下模型。各组分别给予蒸馏水、不同剂量AKCP灌胃,10 d后处死小鼠,检测体质量增率、胸腺指数、脾脏指数、脾淋巴细胞增殖活性和CD3~+/CD4~+/CD8~+亚群、NK细胞杀伤活性、腹腔巨噬细胞吞噬鸡红细胞能力、血清中IL-2和IFN-γ水平。结果 CTX组除NK细胞杀伤活性外的上述各项指标均较对照组明显降低(P<0.05),不同剂量AKCP作用可减轻CTX所致的各项指标下降,其中AKCP高、中剂量组体质量增率、胸腺指数、脾淋巴细胞CD3~+及CD3~+CD8~+亚群、巨噬细胞吞噬率与吞噬指数均较CTX模型组明显升高(P<0.05),AKCP低剂量组脾脏指数、脾淋巴细胞增殖指数以及IL-2和IFN-γ水平显著高于CTX模型组(P<0.05)。结论 AKCP对环磷酰胺致免疫抑制小鼠的免疫功能有保护作用。
Objective To investigate the effects of AKP on the immune function of immunosuppressed mice induced by cyclophosphamide. Methods The mice were randomly divided into 5 groups: blank control group, cyclophosphamide (CTX) model group, AKCP high, medium and low dose groups. In addition to the blank control group, the other 4 groups were given intraperitoneal injection of CTX to establish a mouse model of immunodeficiency. Rats in each group were given distilled water and different doses of AKCP orally. After 10 days, the mice were sacrificed and the growth rate, thymus index, spleen index, proliferative activity of splenic lymphocytes and CD3 + / CD4 + / CD8 + subsets , NK cell killing activity, peritoneal macrophage phagocytosis of chicken erythrocytes, serum IL-2 and IFN-γ levels. Results All the above indexes except CTL group were significantly lower than those of the control group (P <0.05). Different doses of AKCP reduced the decrease of CTX-induced indexes, and the body mass of AKCP high and middle dose groups Compared with CTX model, the phagocytic rate and phagocytic index of macrophages increased significantly (P <0.05), while those of AKCP low dose group , Spleen lymphocyte proliferation index and IL-2 and IFN-γ levels were significantly higher than those in CTX model group (P <0.05). Conclusion AKCP has a protective effect on immune function induced by cyclophosphamide in immunosuppressed mice.
