目的研究体外培养的视网膜Müller细胞中存在的信号传导通路。方法观察从大鼠视网膜分离出的Müller细胞在体外培养过程中的形态变化、生长特性等,利用Protein Pathway Array(PPA)技术检测细胞内146种磷酸化和非磷酸化蛋白质的表达情况,通过信号传导通路分析软件分析Müller细胞内的信号传导通路。结果发现有108种磷酸化和非磷酸化蛋白质在体外培养的大鼠视网膜Müller中表达,这些蛋白质参与了几条重要的信号传导通路,包括XIAP/Apoptosis信号通路,PI3K/AKT信号通路,ILK信号通路,PTEN信号通路,细胞周期:G1/S节点调控信号通路,HIF-1α信号通路,细胞周期:G2/M节点调控信号通路,HGF信号通路,糖尿病信号通路,NFκB信号通路,VEGF信号通路,ERK/MAPK信号通路和胰岛素受体信号通路。结论 Müller细胞中存在多种信号传导通路,这些信号通路相互关联,共同影响Müller细胞生物学特性。
Objective To study the signal transduction pathways existing in cultured retinal Müller cells. Methods The morphological changes and growth characteristics of Müller cells isolated from the retina of rats were observed during the in vitro culture. The expression of 146 phosphorylated and non-phosphorylated proteins in the cells was detected by Protein Pathway Array (PPA) Pathway analysis software analyzes Müller cell signaling pathways. As a result, 108 phosphorylated and non-phosphorylated proteins were expressed in the rat retina Müller cultured in vitro and these proteins are involved in several important signaling pathways including XIAP / Apoptosis signaling, PI3K / AKT signaling, ILK signaling Pathway: PTEN Signal Pathway Cell Cycle: G1 / S Node Regulatory Signal Pathway HIF-1α Signal Pathway Cell Cycle G2 / M Node Regulatory Signal Pathway HGF Signal Pathway Diabetic Signal Pathway NFκB Signal Pathway VEGF Signal Pathway, ERK / MAPK signaling pathway and insulin receptor signaling pathway. Conclusion There are many signal transduction pathways in Müller cells. These signal pathways are related to each other and affect the biological characteristics of Müller cells.
