目的通过基因检测和组织染色的方法,来研究丹参酮IIA磺酸钠对兔耳增生性瘢痕中MMP-1mRNA表达的影响和其对增生性瘢痕的作用。方法①先建立兔耳增生性瘢痕模型,以曲安奈德为阳性对照,分组分次瘢痕基底局部给药,取下瘢痕组织,使用RT-PCR方法检测细胞内金属基质蛋白酶MMP-1基因表达;用苏木素-伊红染色(HE)、苦味酸酸性复红染色法(VG)染色法对成纤维细胞和胶原纤维进行观察。结果 RT-PCR基因检测显示丹参酮及曲安奈德组的MMP-1mRNA的表达显著高于空白对照组;HE染色显示空白对照组中成纤维细胞数量明显高于丹参酮IIA磺酸钠组,丹参酮IIA磺酸钠组与曲安奈德组无明显差异;VG染色显示丹参酮IIA磺酸钠组胶原纤维排列较空白对照组显著整齐,与曲安奈德组无明显差异。结论丹参酮IIA磺酸钠能够通过增强MMP-1mRNA的表达及抑制成纤维细胞的增殖,减少胶原纤维的合成而抑制瘢痕的过度增生。
Objective To investigate the effect of sodium tanshinone IIA sulfonate on the expression of MMP-1mRNA in rabbit ear hypertrophic scars and its effect on hypertrophic scars by gene detection and tissue staining. Methods ① Rabbit ear hypertrophic scar model was established. Triamcinolone acetonide was used as a positive control. The scar base was subcutaneously divided into groups. The scar tissue was removed. The expression of MMP-1 gene was detected by RT-PCR. Hematoxylin and eosin staining (HE) and picric acid complex red staining (VG) staining were used to observe fibroblasts and collagen fibers. Results RT-PCR showed that the expression of MMP-1 mRNA in tanshinone and triamcinolone acetonide groups was significantly higher than that in blank control group. The number of fibroblasts in blank control group was significantly higher than that in tanshinone IIA sulfonate group and tanshinone IIA sulfonate group There was no significant difference between the sodium sulfate group and the triamcinolone acetonide group. VG staining showed that the arrangement of collagen fibers in tanshinone IIA sodium sulfonate group was significantly neat than that in the blank control group, and no significant difference was found between triamcinolone acetonide and triamcinolone acetonide group. Conclusion Sodium tanshinone IIA sulfonate can inhibit the hyperplasia of scar by enhancing the expression of MMP-1 mRNA, inhibiting the proliferation of fibroblasts and reducing the synthesis of collagen fibers.
