目的:检测白细胞介素-23(IL-23)在黑色素瘤中的表达并探讨IL-23对黑色素瘤细胞双向分化及增殖、侵袭能力的影响。方法:将IL-23降表达质粒转染至黑色素瘤细胞系MUM-2B中,使MUM-2B中IL-23外源性降表达,观察IL-23转染前、后MUM-2B细胞形态学改变;MTT、细胞划痕和Transwell侵袭实验检测对增殖、迁移及侵袭能力的影响;Western blot检测转染前、后MUM-2B中IL-23和相关蛋白(Vimentin、Snail、N-cadherin和MMP-2)表达变化情况;免疫荧光检测IL-23、Vimentin和MMP-2表达部位,明胶酶谱检测MMP-2、MMP-9活性。结果:转染后MUM-2B的细胞形态发生明显改变。细胞的增殖、迁移及侵袭能力显著减弱。IL-23降表达,N-cadherin、Vimentin、NF-κβ、Snail和MMP-2表达明显下调,MMP-2、MMP-9活性显著降低。结论:IL-23可促进黑色素瘤细胞向间叶样表型转变,并促进黑色素瘤细胞的增殖、迁移和侵袭。
Objective: To detect the expression of interleukin-23 (IL-23) in melanoma and to explore the effect of IL-23 on the bi-directional differentiation, proliferation and invasion of melanoma cells. Methods: The IL-23 down-expression plasmid was transfected into the melanoma cell line MUM-2B, and the expression of IL-23 in MUM-2B was induced exogenously. The morphology of MUM-2B cells before and after IL-23 transfection The effects of MTT, cell scratch and Transwell invasion on the proliferation, migration and invasion of MUM-2B cells were detected by Western blot. The expressions of IL-23 and related proteins (Vimentin, Snail, N-cadherin and MMP -2). The expression of IL-23, Vimentin and MMP-2 were detected by immunofluorescence. The activity of MMP-2 and MMP-9 was detected by gelatin zymography. Results: The morphological changes of MUM-2B cells after transfection were significantly changed. Cell proliferation, migration and invasive ability was significantly weakened. The expression of IL-23 was down-regulated while the expressions of N-cadherin, Vimentin, NF-κβ, Snail and MMP-2 were down-regulated while the activities of MMP-2 and MMP- Conclusion: IL-23 can promote melanoma cells to mesenchymal phenotype and promote melanoma cell proliferation, migration and invasion.
