目的建立一种可以同时检测人类A型流感病毒,B型流感病毒及A型流感病毒H1、H3亚型的多重PCR技术,并与单重PCR技术进行比较与应用。方法根据人类流感病毒的特异性基因设计并合成多对特异性引物,建立可同时扩增出人类流感病毒亚型特异性片段的多重PCR的方法。在对病毒的检测中与单重PCR进行特异性和灵敏度的比较。结果单重PCR对A型流感病毒检测灵敏度是44.68%(21/47),特异度是99.70%(686/688);对B型流感病毒检测的灵敏度是18.18%(2/11),特异度是97.38%(670/688)。多重PCR法对A型流感病毒检测灵敏度是44.68%(21/47),特异度是97.38%(670/688);对B型流感病毒检测的灵敏度是18.18%(2/11),特异度是97.67%(672/688)。结论本研究建立多重PCR可以准确检测流感病毒,而且所用时间短,可以快速得到检测结果。
Objective To establish a multiplex PCR technique that can simultaneously detect human influenza A virus, influenza B virus and H1 subtype of influenza A virus, and to compare with single PCR. Methods Based on the specific genes of human influenza virus, multiple pairs of specific primers were designed and synthesized, and a multiplex PCR method was developed to amplify specific fragments of human influenza virus subtypes simultaneously. Specificity and sensitivity were compared with single PCR in the detection of the virus. Results The sensitivity and specificity of single PCR for detecting influenza A virus were 44.68% (21/47) and 99.70% (686/688), respectively. The sensitivity of detection to influenza A virus was 18.18% (2/11) Is 97.38% (670/688). The sensitivity of multiplex PCR was 44.68% (21/47) for influenza A virus and 97.38% (670/688) for influenza A virus, 18.18% (2/11) for influenza B virus, and the specificity was 97.67% (672/688). Conclusion This study established a multiplex PCR can accurately detect influenza virus, and the use of time is short, you can quickly get the test results.
