目的 :探讨更昔洛韦 (GCV)对人白血病细胞的抗增殖和诱导分化作用。方法 :GCV加入K5 6 2细胞培养 4d ,测定其活细胞数目、克隆形成率、联苯胺染色阳性率 ,观察光镜形态、组织化学染色 ,并进行流式细胞分析、端粒酶检测。结果 :在GCV的作用下 ,K5 6 2细胞的增殖受抑 ,生长分数降低 ,并且向具有合成血红蛋白能力的细胞分化。结论 :GCV对K5 6 2细胞具有抑制增殖和诱导分化作用 ,该结果为慢性粒细胞白血病 (CML)的治疗探索新的途径。
Objective: To investigate the anti-proliferation and differentiation of human leukemia cells treated with ganciclovir (GCV). Methods: The number of viable cells, the rate of colony formation, the positive rate of benzidine staining, the morphology of light microscope and the histochemical staining were observed after the cells were cultured with K562 cells for 4 days. The expression of telomerase was detected by flow cytometry. Results: Under the action of GCV, the proliferation of K562 cells was inhibited, the growth fraction was decreased, and the cells with the ability of synthesizing hemoglobin were differentiated. CONCLUSION: GCV can inhibit proliferation and induce differentiation of K562 cells. This result provides a new approach for the treatment of chronic myelogenous leukemia (CML).
