以10对SSR玉米核心荧光引物为研究对象,探讨了多重PCR组合的建立,通过对两重PCR组合的优化,找到了适于两重、三重及四重PCR优化的方法;通过正交设计,快速找到了适于六重PCR的扩增体系,并且应用六重PCR体系成功构建了1个七重PCR。
Ten pairs of SSR maize core fluorescent primers were used as research objects to explore the establishment of multiple PCR combinations. By optimizing the combination of the two PCRs, two methods for the optimization of two, three and four-fold PCR were found. By orthogonal design, The amplification system suitable for six-fold PCR was rapidly found, and a seven-fold PCR was successfully constructed by using the six-fold PCR system.
