目的:探讨人脐带间充质干细胞(HUC-MSCs)条件培养基对舌鳞癌Cal-27细胞生物学特性的影响。方法:培养HUC-MSCs和Cal-27细胞,倒置显微镜下观察细胞形态;将HUC-MSCs条件培养基与Cal-27细胞体外间接共培养,CCK-8法检测Cal-27细胞的增殖活性,绘制生长曲线;通过Western blot法检测HUC-MSCs条件培养基对Cal-27细胞凋亡蛋白及Wnt/β-catenin信号通路关键蛋白的表达。结果:生长曲线结果显示,HUC-MSCs条件培养基可抑制Cal-27细胞增殖,并呈一定的浓度依赖性,这些变化伴随Cal-27细胞内PARP-1表达上调,Bcl-2、β-catenin、cMyc表达下调。结论:HUC-MSCs条件培养基可抑制Cal-27细胞增殖并促使其凋亡,这可能与细胞内的Wnt/β-catenin信号通路被抑制有关。
Objective: To investigate the effect of human umbilical cord mesenchymal stem cells (HUC-MSCs) conditioned medium on the biological characteristics of tongue squamous cell carcinoma Cal-27 cells. Methods: HUC-MSCs and Cal-27 cells were cultured in vitro. Morphology of HUC-MSCs was observed under inverted microscope. Indirect co-culture of HUC-MSCs conditioned medium and Cal-27 cells was performed. The proliferation activity of Cal-27 cells was detected by CCK- The growth curve of HUC-MSCs was detected by Western blot. The expressions of apoptosis protein and key proteins of Wnt / β-catenin signaling pathway were detected by Western blot. Results: The results of growth curve showed that HUC-MSCs conditioned medium could inhibit the proliferation of Cal-27 cells in a concentration-dependent manner. These changes were associated with up-regulation of PARP-1 expression, Bcl-2 and β-catenin , CMyc expression down regulation. CONCLUSION: HUC-MSCs conditioned medium can inhibit the proliferation and induce apoptosis of Cal-27 cells, which may be related to the inhibition of intracellular Wnt / β-catenin signaling pathway.
