目的研究保肾方Ⅱ号对糖尿病肾病大鼠细胞外基质(ECM)的影响。方法以40只大鼠由链脲佐菌素(STZ)诱导造成糖尿病模型,3d后随机分为5组,于第16周检测各组大鼠的有关指标,于16周采用免疫组织化学法检测肾皮质基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶组织抑制因子-2(TIMP-2)的表达情况。结果保肾方Ⅱ号大剂量组MMP-2与TIMP-2的表达高于保肾方Ⅱ号小剂量组、洛汀新对照组、空白模型对照组,低于正常组。结论保肾方Ⅱ号对糖尿病肾病大鼠肾功能具有保护作用,可降低血糖,可上调肾组织MMP-2、TIMP-2的表达,调节MMP-2/TIMP-2的比例失衡,促进ECM成分的降解,减缓糖尿病肾病肾小球纤维化的进程。
Objective To study the effect of Baoshen Formula II on extracellular matrix (ECM) in diabetic nephropathy rats. Methods Diabetic models were induced by streptozotocin (STZ) in 40 rats. After 3 days, they were randomly divided into 5 groups. The relevant indexes of each group of rats were detected at the 16th week and examined by immunohistochemical method at 16 weeks. The expression of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in renal cortex. Results The expression of MMP-2 and TIMP-2 in Baoshenfang II high-dose group was higher than that in Baoshenfang II low-dose group, Lotensin new control group and blank model control group, but lower than the normal group. Conclusion Baoshenfang II has a protective effect on renal function in rats with diabetic nephropathy, can reduce blood glucose, can upregulate the expression of MMP-2 and TIMP-2 in renal tissue, regulate the imbalance of MMP-2/TIMP-2, and promote ECM composition. The degradation slows the progression of glomerular fibrosis in diabetic nephropathy.
