目的研究缺氧诱导因子-1α(HIF-1α)和肿瘤细胞增殖抗原Ki67在乳腺癌及癌旁组织及良性肿瘤组织中的表达情况;分析HIF-1α和Ki67之间的相关性,初步探讨HIF-1α对乳腺癌细胞增殖的影响。方法取2008至2010年在桂林医学院附属医院手术治疗后存档并留有病理标本的70例乳腺癌、10例乳腺良性肿瘤组织标本,采用快捷免疫组化S-P法检测乳腺癌组织及其癌旁组织和良性肿瘤组织中HIF-1α和Ki67的表达情况,研究它们与临床病理特征的关系,进一步探讨缺氧对乳腺癌细胞增殖的影响。结果 70例乳腺癌组织中HIF-1α和Ki67的阳性表达率分别为78.6%(55/70)和88.6%(62/70),其癌旁组织中HIF-1α和Ki67的阳性表达率分别为45.7%(32/70)和11.4%(8/70),10例乳腺良性肿瘤组织中HIF-1α的阳性表达率为0(0/10),Ki67的表达率均为阴性,差异有统计学意义(P<0.01);其表达与临床分期及腋淋巴结转移密切相关(P<0.05),但与患者年龄、绝经状况、肿块大小、激素受体无明显相关,在乳腺癌组织中HIF-1α与Ki67的表达强度呈正相关(P<0.05),在癌旁组织及良性肿瘤组织未见明显相关(P>0.05)。结论乳腺癌组织中HIF-1α高表达,与淋巴结转移及组织恶性程度相关,提示HIF-1α可能成为预测乳腺癌侵袭、转移和预后的重要指标;HIF-1α和Ki67二者表达强度呈明显正相关,提示缺氧可促进乳腺癌细胞增殖,但确切作用机制有待进一步研究。
Objective To investigate the expression of hypoxia-inducible factor-1α (HIF-1α) and Ki67 in breast cancer, paracancerous tissues and benign tumor tissues. To analyze the correlation between HIF-1α and Ki67, -1α on the proliferation of breast cancer cells. Methods 70 cases of breast cancer and 10 cases of benign breast tumor were archived with pathological specimens after operation in Affiliated Hospital of Guilin Medical College from 2008 to 2010. The fast immunohistochemical SP method was used to detect breast cancer tissues and their adjacent tissues HIF-1α and Ki67 in tissues and benign tumor tissues, and to study the relationship between HIF-1αand Ki67 in clinicopathological features and further explore the effect of hypoxia on the proliferation of breast cancer cells. Results The positive expression rates of HIF-1α and Ki67 in 70 cases of breast cancer were 78.6% (55/70) and 88.6% (62/70), respectively. The positive rates of HIF-1α and Ki67 in adjacent tissues were 45.7% (32/70) and 11.4% (8/70) respectively. The positive expression rate of HIF-1α in 10 cases of benign breast tumor was 0 (0/10), the expression rate of Ki67 was negative, the difference was statistically significant (P <0.01). The expression of HIF-1α was closely related to clinical stage and axillary lymph node metastasis (P <0.05), but not to age, menopausal status, tumor size and hormone receptor. There was a positive correlation between the expression of Ki67 and the intensity of Ki67 (P <0.05). There was no significant correlation between the expression of Ki67 and adjacent tissues (P> 0.05). Conclusions HIF-1α overexpression in breast cancer is correlated with lymph node metastasis and histological grade, suggesting that HIF-1α may be an important index for predicting the invasion, metastasis and prognosis of breast cancer. The expression of HIF-1α and Ki67 are significantly positive Related, suggesting that hypoxia can promote breast cancer cell proliferation, but the exact mechanism needs further study.
