目的研究糖皮质激素(glucocorticoids)对牙周膜细胞表达骨保护因子(osteoprotegerin,OPG)的影响。方法临床收集青少年患者的新鲜正畸拔除前磨牙,刮取牙周膜组织进行细胞培养。采用有限稀释法获得牙周膜细胞克隆,并进行细胞克隆鉴定。培养基中加入地塞米松(dexamethasone,DEX),MTT法检测牙周膜细胞增殖水平,夹心酶联免疫吸附法测定培养上清液中OPG含量。结果 DEX质量浓度在40 ng/mL以上时显著降低细胞对OPG的表达量(P<0.001);DEX对细胞增殖没有明显影响(P>0.05)。结论糖皮质激素可以通过抑制牙周膜细胞对OPG的表达提高牙周骨质吸收活性。
Objective To study the effect of glucocorticoids on the expression of osteoprotegerin (OPG) in periodontal ligament cells. Methods Fresh orthodontic removal premolar teeth were collected from adolescent patients and the periodontal ligament tissues were scraped for cell culture. Periodontal ligament cell clones were obtained by limiting dilution method, and cell clones were identified. The dexamethasone (DEX) was added to the culture medium, the proliferation of periodontal ligament cells was detected by MTT method, and the content of OPG in culture supernatant was determined by sandwich enzyme-linked immunosorbent assay. Results The DEX concentration of 40 ng / mL or more significantly reduced the expression of OPG in cells (P <0.001). DEX had no significant effect on cell proliferation (P> 0.05). Conclusion Glucocorticoids can improve the periodontal bone resorption activity by inhibiting the expression of OPG in periodontal ligament cells.
