目的建立高效毛细管电泳二极管阵列检测法(HPCE-DAD)同时测定地黄饮片中梓醇、桃叶珊瑚苷、栀子苷、毛蕊花糖苷和肉桂酸含量的方法。方法以60 mmol.L1硼砂缓冲液(5%甲醇,pH 9.5)为电泳介质,未涂渍标准熔融石英毛细管(75μm×64.5 cm,有效长度56 cm)为分离通道,分离电压为20 kV,检测波长为210 nm,毛细管温度为25℃,压力进样为5 kPa×6 s。结果 5种指标成分的浓度与峰面积的线性关系良好(r>0.997 8);加样回收率为97.63%~103.03%。用此法测定了地黄饮片中5种指标成分的含量,得到满意结果。结论该方法简单、准确,重复性较好,可用于地黄饮片内在质量的评价和控制。
OBJECTIVE To establish a method for the simultaneous determination of catalpol, aucubin, geniposide, verbascoside and cinnamic acid in Rehmanniae Pieces by high performance capillary electrophoresis with diode array detection (HPCE-DAD). Methods 60 mmol·L1 borax buffer (5% methanol, pH 9.5) was used as the electrophoresis medium. The standard fused silica capillary (75 μm × 64.5 cm, effective length 56 cm) was used as the separation channel. The separation voltage was 20 kV. The wavelength is 210 nm, the capillary temperature is 25 ℃ and the pressure is 5 kPa × 6 s. Results The linear relationship between the concentration of the five index components and the peak area was good (r> 0.997 8). The recoveries of samples were 97.63% -103.03%. The method was used to determine the contents of five index components in Rehmanniae Pieces, and satisfactory results were obtained. Conclusion The method is simple, accurate and reproducible. It can be used to evaluate and control the intrinsic quality of Decoction Pieces.
