目的:研究疏肝健脾活血方对肝纤维化过程中大鼠TGF-β1/Smad信号通路的影响。方法:采用CCl4诱导大鼠肝纤维化模型。通过苏木精-伊红(HE)染色和Masson染色作肝组织病理学观察,比较纤维化程度,并用ELISA法检测血清中Ⅰ、Ⅲ、Ⅳ型胶原含量的变化;Western Blot检测TGF-β1和p-Smad2/3的蛋白表达水平;逆转录聚合酶链反应(RT-PCR)法检测大鼠肝组织中TGF-β1、Samd2、Samd3、Samd4及Samd7的m RNA表达水平。结果:与模型组比较,疏肝健脾活血方各剂量组肝纤维化程度显著降低,肝组织中TGF-β1和p-Smad2/3的蛋白表达量及Samd2、Samd3和Samd4的m RNA表达量显著降低(P<0.01,P<0.05),Samd7 m RNA表达量显著上升(P<0.01,P<0.05)。结论:疏肝健脾活血方对大鼠肝纤维化具有显著干预作用,其作用机制可能通过抑制TGF-β1、Samd2、Samd3、Samd4、p-Smad2/3和上调Smad7的表达,以减少信号的后续转导,进而抑制肝纤维化的进展。
Objective: To investigate the effect of Shugan Jianpi Huoxue Recipe on TGF-β1 / Smad signaling pathway in rats with liver fibrosis. Methods: CCl4 induced hepatic fibrosis in rats. The hepatic histopathological changes were observed by hematoxylin-eosin (HE) staining and Masson staining, and the changes of collagen typeⅠ, Ⅲ and Ⅳ were detected by ELISA. The levels of TGF-β1 and The expression of p-Smad2 / 3 was detected by RT-PCR. The mRNA expression of TGF-β1, Samd2, Samd3, Samd4 and Samd7 were detected by reverse transcription polymerase chain reaction (RT-PCR) Results: Compared with the model group, the liver fibrosis of Shugan Jianpi Huoxue Decoction decreased significantly. The protein expression of TGF-β1 and p-Smad2 / 3 and the mRNA expression of Samd2, Samd3 and Samd4 (P <0.01, P <0.05). The expression of Samd7 mRNA increased significantly (P <0.01, P <0.05). Conclusion: Shugan Jianpi Huoxue Recipe has significant intervention on hepatic fibrosis in rats, and its mechanism may be through inhibiting TGF-β1, Samd2, Samd3, Samd4, p-Smad2 / 3 and up-regulating Smad7 expression, Follow-up transduction, thereby inhibiting the progress of liver fibrosis.
